Often times, yeah. If you've never fired a gun before, I'd stay away from the larger caliber handguns at first. Until you know what you're getting into. Firing a .44 Magnum is going to have way more kick than a lot of people expect. And don't even think of picking up a Desert Eagle like they love in movies and video games. That thing can dislocate your arm if you're not properly handling it.

I'd recommend your first time shooting be something with less recoil and power. .22 or a 9mm cartridge would be a good first one to try, and then moving to a heavier handgun caliber once you get at least a bit of a feel for it. It shouldn't take long, though.
